click hereEFFECT OF 2,4-D ON CALLUS INDUCTION AND PLANT REGENERATION IN THREE INDEGENOUS AROMATIC RICE CULTIVARS OF BANGLADESH
The present
investigation was carried out to find out the most suitable concentrations of
2, 4- D and the potentiality of selected aromatic local rice cultivars for
maximum callus induction and plant regeneration. Matured dehusked seeds of
three local aromatic rice cultivars viz Raniselute,
Kachda and Chiniatap were cultured on MS medium supplemented with different
concentrations of 2,4-D at 0.5, 1.0, 1.5, 2.0, 2.5, 3.0 mgL-1.
MS media supplemented with 2, 4-D at 1.0 mg L-1 performed better for
callus induction in Chiniatap (88%) and in Raniselute (76%) at 2.0 mg L-12,4-D, Kachda
performed better (76%) than other concentration. The cultivar
Kachda showed comparatively better performance in morphogenesis (90.05%) than
Raniselute (84.89 %) and Chiniatap (71.46 %). MS media supplemented with 2,4-D
at 2.0 mgL-1 performed the best for morphogenesis (90.9%) in
Raniselute and ( 90%) in Chiniatap. The cultivar Kachda showed comparatively
better performance (23.67%) in plantlet regeneration than Raniselute (14.69%)
and Chiniatap (7%). MS media supplemented with 2,4-D at 1.0 mgL-1
performed the best in Raniselute (20.83%) and in Chiniatap (13.33%) for plantlet regeneration. But in Kachda , it
was best (31.58%) in 2.0mgL-1. The rate of survivability of Kachda (32%) was higher than Raniselute (9%)
and Chiniatap (8%). In this respect, the cultivar Kachda and MS medium
supplemented with 2, 4-D at 1.0 mgL-1and 2.0 mgL-1may be
recommended for the further study of somaclonal variation and for genetic
engineering studies.
